Getting to Samarkand
From Tashkent, the Afrosiyob high-speed train takes about 2 hours (economy about 311,000 UZS, business about 455,000 UZS). Onward to Bukhara it is about 1 hour 40 minutes (economy about 225,000 UZS). Timetables and days of operation vary, so ask the guide for your date; see also the Tashkent–Samarkand train guide.
Go to the Registan at sunset or in the evening when it is lit, and to Shah-i-Zinda early, before tour groups. See Samarkand in one day.
Do I need a live guide in Samarkand?
You can see Samarkand without one: the main monuments are well signed. A live guide helps with the history of the Registan and Shah-i-Zinda. For a self-guided option see audio tours.
